Research / Synopsis

Our research interests focus on fatty acid and neutral lipid (triglyceride) metabolism in the yeast Saccharomyces cerevisiae. We are interested in the regulation of fatty acid synthesis (acetyl-CoA carboxylase) and the regulatory role of fatty acids in cellular metabolism. Related to this problem is the question of fatty acid storage in the form of triglycerides, and their mobilization by lipases, which is the focus of our research in the framework of the GOLD - Genomics of Lipid-associated Disorders project. The SFB LIPOTOX focuses on the detrimental and ultimately toxic effects of de-regulated lipid metabolism on cellular physiology and function. These studies involve biochemical and analytical (mass spectrometry), molecular, and cell biological techniques as well as genomics, and bioinformatics approaches, making full use of the experimental potential of yeast for biomolecular and biomedical research. Cytological studies in yeast and other cell types involve video and confocal laser scanning microscopy, which also implements CARS, SHG and other multi photon applications; these technologies are developed into high-throughput high-content imaging methods, for mutant and drug screens (project mYeasty). We have established a yeast protein localization database, YPL.db, that displays microscopic localization patterns of proteins involved in lipid metabolism. We also employ various yeast species as hosts for heterologous gene expression, for functional studies in vivo and large scale protein production.
Special emphasis of our work is on problems related to lipid-associated disorders and is thus of high significance for biomedical research in humans.
